I have a Dell 5100cn that was reading 'Replace Transfer Roller'. The machine would still print, albeit poorly. I replaced the Transfer Roller and the print was somewhat better but with the same error message. I issued the command to clean the magenta color (as I recall) and at the end of the cycle the machine still had the error 'Replace Transfer Roller' but now does not print at all.
Advice for troubleshooting this unit and resolving the issue is hard to find. The only solution that I have found resolves the issue by replacing the Machine Control Unit. This is a costly repair if it does not solve the problem.
What can I do to narrow the solution to a the correct part?
The transfer roller on this machine comes as an assembly consisting of a waste toner tank and a transfer roller. The message you are getting pops up when this tank is full, regardless of the transfer roller's useable life. There is a window on the back that when blocked by toner, cuts off an optical sensor to tell the machine that the waste toner is full. Just unclip the roller from the side, pop it out, and dump the toner out. Careful though, it's a messy job and you might want to wrap the assembly inside a trash bag while emptying it out.
I took the transfer roller out of the printer and subjected it to compressed air from my compressor in the garage. Do this outside, as you'll find that toner streams out all over the place. I was able to force air into the roller in certain areas that appeared to force the plastic out enough for the toner trapped in the transfer roller to escape. I think that, in my case, magenta toner got oversupplied and filled up the transfer roller, as that is what came out when I forced compressed air through it. I also blew out the inside of the printer to make sure any optical sensors were not covered with toner. Had to power the printer off and on after replacing the transfer roller to get rid of the error message.
Right now the printer seems to be working well.

I have reset the print count. The problem was resolved by cleaning the optical sensor that reads levels of the transfer roller waste tank. I used canned air to blow out any toner particles and the machine began to print again. But the print is poor. A test print of pur black shows a light area, maroon in color, on one side of the page and has two horizontal lines approximately 8 px high, also maroon in color. A print of a mostly black image show horrible maroon discoloration on one side of the print and smudges throughout. What's next?
Although this post is ancient, I am glad I stumbled across it because you are absolutely correct. Once I popped the white tab over and pulled the transfer roller out, I shook the cartridge inside a garbage bag and it prints just as good as day one now! Thanks!
