I was also occuring from same problem. Do the following steps:-
1. Right click on my computer icon and open manage.
2. Then open disk management.
3. There u see ur pen drive.
4. Right click on ur pen drive and click on format.
5. After successful formating restart ur computer.
6. Ur pen drive wil work again.

If you are unable to open it ,there might be a virus in you flash drive which is causing problem . Solution: Select the flash drive and right click .now select autoplay, a dialogue box will appear ,now select open to open it.. if you are unable to do this . open my computer ,in the address bar ,click on the downn area button in address bar and select flash drive to open .if this also fails you have to format the flash drive ..
